Dr. Prasanna Ramani did his master’s degree in Chemistry from Bharathidasan University, India and Ph. D. in Chemical Sciences from University of Milan, Italy.

His doctoral thesis work was focused on Synthesis/labeling of Modified Peptide Nucleic Acids (artificial DNA) and their Analytical Properties. Dr. Ramani worked on polymer chemistry, total synthesis of natural products and functionalization of CNTs during his tenure as a postdoctoral researcher at Center for bio-molecular Interdisciplinary Studies and Industrial applications (CISI), University of Milan, Italy. His postdoctoral research was supported by UNESCO grant from University of Trieste, Italy.

Before his doctoral studies, Dr. Ramani worked for three years as Senior Chemist at Orchid Chemicals and Pharmaceuticals Ltd., Chennai, India. He has organized, participated and presented in various meetings and workshops around India and Europe.

Dr. Ramani is also guiding a number of undergraduate & postgraduate student projects and Ph. D. research scholars. He currently researches on synthesis of biologically interesting molecules and natural product chemistry.

The research team targets on the synthesis of new molecules that can target cancer. Cancer results from mutation of certain genes that allow the cells to begin their uncontrolled growth. These mutations are inherited or acquired. The group members intend to find a possible solution towards cancer free world. The following are our Research interests:

  • Peptide Nucleic Acids:

The remarkable binding properties of ‘Peptide Nucleic Acids’ (PNA), the so called artificial DNAs, have spawned a rapidly expanding new field of research and their applications as therapeutics, DNA diagnostics, tools in biotechnology and much more. PNA is linked to various scientific disciplines, few includes medicinal chemistry, biochemistry, molecular biology, immunology and drug development. Our focus is to improve the solubility and sensitivity to detect complementary natural nucleobases. 

  • Medicinal Chemistry:

Endangered Plants:

Herbal medicines are assumed to be of great importance in the primary health care of individual and communities. The World Health Organization has estimated that 80% of the population of developing countries still relies on traditional medicines, mostly plant drugs, for the primary health care needs. The high degree of efficacy and safety with herbal medicines make them more acceptable compared to other therapeutic invention. Plant-based traditional knowledge has become a recognized tool in search for new sources of drugs and neutraceuticals. Our basic aim is to identify the plants which are on the verge of extinction and find the medicinal value of that plant, which can be used by us in our day today life.

Anti-Cancer molecules:

Alkaloids are a class of naturally occurring organic nitrogen containing compounds. Typical alkaloids are derived from plant sources, they are basic, they contain one or more nitrogen atoms (usually in a heterocyclic ring) and they usually have a marked physiological action on man or other animals. Alkaloids have potent anticancer activity against various cancers. Our intentions are to synthesis some alkaloids and study their biological properties.

  • Combinatorial chemistry:

In this, our research has been focused on the design and synthesis of small molecules towards human carcinoma. This work is in collaboration with Dr. Krishnan Namboori of Computational Chemistry Group. Molecules generated through computer-assisted molecular design will be screened for synthetic feasibility. Selected molecules will be synthesized in our laboratory and subjected to biological evaluation.

  • Drug Delivery:

 Sustained Drug Delivery refers to approaches, formulations, technologies, and systems for transporting a pharmaceutical compound in human system as needed to safely achieve its desired therapeutic effect. This can be attained by maintaining a constant drug concentration for a specific period of time with minimum side effect. Most pharmaceutical drugs in the market are encapsulated with gelatin, which is manufactured from the collagen of animal skin or bone. The target in this research is to find an alternative means of encapsulating those drugs for sustained release.

A series of new 4-aryl-4H-chromene-3-carbonitrile derivatives were obtained by one-pot synthesis using substituted benzaldehydes, malononitrile, and substituted phenols. All the synthesized compounds (1a, 1b, 1c, 1d, 1e) were screened in vitro for antioxidant and anticancer activities. Compounds 1c, 1d, 1e showed significant antioxidant activity in nitric oxide free radical scavenging method while compounds 1c and 1e showed significant activity in hydrogen peroxide free radical scavenging method. The other compounds showed significant to moderate activities in both the methods in comparison with ascorbic acid and butylated hydroxytoluene as standards. Compounds 1c, 1d, 1e exhibited good anticancer activity, using Michigan Cancer Foundation-7 (MCF-7) cell line, compared with those of other synthesized compounds. © 2015 HeteroCorporation.

Benzo[c]phenanthridine alkaloid, 7-hydroxynitidine, was synthesized from readily available 2-benzyloxy-6-bromo-3,4-dimethoxybenzaldehyde 5 and napthylamine 6 using reductive amination followed by radical cyclization in eight steps. This method is highly efficient and better way to synthesize fully aromatized benzo[c]phenanthridine compounds.

Carboxy-terminated polyvinylpyrrolidin-2-one (PVP) has been used as a new water-soluble and biocompatible polymeric support for a series of ferrocene labeled amino acid and peptide nucleic acid (PNA) monomer derivatives 4–7. The organometallic polymer-conjugates thus obtained are new and potentially useful as water-soluble electrochemically active probes for biomolecules. In view of such application, their electrochemical activity has been evaluated and has proved very high notwithstanding the complexity and bulkiness of the molecule, affording detection limits down to 10−8 M in the aqueous medium.

A new chiral ferrocene-labelled tyrosine PNA monomer 1 has been synthesised in good yield in both racemic and enantiomerically pure forms. It is suitable for insertion in various positions of PNA oligomers, a possibility that has been preliminarily demonstrated by synthesising the dimer 16. Moreover, in view of possible applications to nucleic acid detection, a preliminary voltammetric investigation on the electrochemical activity of monomer 1 and its synthetic precursors has been carried out in DMF. It appears that, despite the bulkiness of the PNA monomer backbone, its insertion on the ferrocene group only moderately lowers the latter’s diffusion coefficients and peak currents, thus affording voltammetric detection limits in the order of 10−6–10−7 M.
